FAIN, JOHN N., MARK D. JACOBS, AND YVONNE C. CLEMENT-, CORMIER. Interrelationship of cyclic AMP, Zipolysis, and respiration in brown fat cells. Am. J. Physiol. 224(Z): 346-351. 1973.-The marked stimulation of oxygen consumption in brown fat cells seen after the addition of norepinephrine is thought to be secondary to the stimulation of adenylate cyclase and result from a K+-dependent uncoupling action of free fatty acids released as a result of activation of lipolysis by cyclic AMP. The present studies indicate that there was a good correlation between the ability of catecholamines and serotonin (5-hydroxytryptamine) to affect cyclic AMP accumulation, lipolysis, and respiration. Propranolol, a beta-adrenergic antagonist, blocked the effects of both serotonin and norepinephrine. High concentrations of ouabain reduced the stimulation of cyclic AMP accumulation, lipolysis, and oxygen consumption by catecholamines but did not inhibit the effects of theophylline on lipolysis or cyclic AMP accumulation. The stimulation of respiration by added fatty acids was also unaffected by ouabain. Ouabain increased glucose conversion to carbon dioxide and decreased lactate accumulation.

Metabolic relationships between lipolysis and respiration in rat brown adipocytes. The role of long chain fatty acids as regulators of mitochondrial respiration and feedback inhibitors of lipolysis.

The calorigenic action of norepinephrine in isolated brown adipocytes was selectively mimicked by theophylline, dibutyryl cyclic AMP, and the principal fatty acids known to be present in the acyl moieties of brown adipose tissue triglycerides (palmitic, oleic, and linoleic acids). The stimulatory effects of fatty acids were entirely reversible, occurred at physiological concentrations, and were...

Preparation of hormone-sensitive adipocytes from foetal rat brown adipose tissue.

A collagenase-dispersion method involving Percoll gradient separation is described for the production of adipocytes from foetal rat brown adipose tissue. The cells produced are hormone-sensitive as judged by cAMP accumulation and lipolysis.

Relationship between the tissue level of cyclic AMP and the fat cell size of human adipose tissue.

The relationship between mean fat cell size, maximal tissue cyclic AMP concentration, and glycerol release was investigated in human subcutaneous adipose tissue incubated in vitro with or without isoprenaline or noradrenaline added at maximal effective concentrations. Basal and stimulated glycerol release and cyclic AMP concentration were each related to the fat cell size. Whether or not the ph...

Beige Adipocytes Are a Distinct Type of Thermogenic Fat Cell in Mouse and Human

Brown fat generates heat via the mitochondrial uncoupling protein UCP1, defending against hypothermia and obesity. Recent data suggest that there are two distinct types of brown fat: classical brown fat derived from a myf-5 cellular lineage and UCP1-positive cells that emerge in white fat from a non-myf-5 lineage. Here, we report the isolation of "beige" cells from murine white fat depots. Beig...
